Manjrekar has acted in a number of movies, some of which he has directed. He made his screen debut as a leprosy patient in the Door darshan Marathi series Kshitij. His performance in the 2002 movie Kaante won him praise for his acting abilities. He went on to play antagonistic characters in the films Arrambam (2013), Okkadunnadu (2007), and Slumdog Millionaire (2008), as well as negative roles in Telugu and Tamil. Me Shivajiraje Bhosale Boltoy, a Marathi film, featured him as Chatrapati Shivaji Maharaj.
