Tripathi got small roles in films like Omkara (2006), Raavan (2010), and Agneepath (2012), and a supporting part in the TV show Powder (2010) after studying acting at the National School of Drama.

Tripathi was the youngest of four children born to Pandit Benares Tiwari and Hemwanti Tiwari, in Belsand village, Barauli, in the Gopalganj District of the Indian state of Bihar. She was born on September 5, possibly in 1975.

In addition to farming, his father was a Hindu priest. When he was in the ninth grade, Tripathi legally changed his last name from Tiwari to Tripathi. He primarily portrayed negative characters in the early years of his career, which led to his association with gangster personas. Pankaj Tripathi was nominated for a Filmfare OTT Award for Best Actor in a Web Original Film for his performance in the biographical comedy Kaagaz (2021) on ZEE5.

Pankaj Tripathi co-starred with Akshay Kumar and Yami Gautam in the satirical comedy-drama OMG 2 and the comedy Fukrey 3. Next on his agenda is a portrayal of Atal Bihari Vajpayee, the former prime minister of India, in the biographical play Main Atal Hoon. The movie is scheduled to open on January 19, 2024.
